In addition, Microsoft has also developed a Malicious Software Removal Tool. It won't eradicate spyware on your system, but it's handy for rooting out particularly hard to kill worms, such as Blaster and Sasser. Add it to your Windows toolkit.
Also check out Crap Cleaner which will clean out your internet, OS and apps of various logs, temp files, cache, etc.
Go to Sysinternals, click on the security link and download the RootkitRevealer application which will check your system for Rootkits.
